Can you recommend a reputable guide who could take 2 adults from Palermo to Trapani for a 1 day excursion in late September?

What do you want to see in Trapani?
I spent a few nights there recently, and it was a great base for visiting Segesta, Erice, and the Egadi Islands.
You can certainly visit Segesta, Erice, Cefalù, and Monreale from Palermo.

We are going to be in Palermo for 3 days. I would like spend one of those days visiting Erice and Trapani but do not want to drive. That's why we are looking for a reputable tour guide.

My apologies... I realize that my reply was not helpful to you.
Perhaps your hotel staff may be able to recommend a reputable tour for you.
If you have time, you can spend a few days in Trapani and take day trips to Erice & Segesta from there via public transportation.
Are you planning to visit Monreale?
You can easily get there via public bus.
Cefalù is a nice seaside town and can be reached by train from Palermo.
