We will be in Prague in September and want to go to Karlovy Vary for the day. We don't want a guide, just need to get there and back. I have read that the RegioJet Bus is the best way to get there. Has anyone done that? Or are there other options for getting there? There are two of us, myself and my 26 year old son.

The RegioJet Bus is more than 1 hour faster than the train and service is frequent and cost is reasonable. Cost is reasonable as well at 6.40 EUR per person each way.

Karlovy Vary is one of my favorite places in the world. Yes, the bus is the fastest public transit from Prague. It’s serviced by both RegioJet as well as FlixBus. Both arrive/depart from the train station and offer comfortable buses. If you’re returning to Prague on a Sunday, make sure you pre-book your return ticket as the buses book up and you maybe out of luck. That happened to me. I had to take the train which is almost two hour longer as it goes thru Usti nad Labem and many mountains. An alternate would be to Uber it from Prague. My train from Krakow was delayed for track work causing me to miss the last bus of the night to KV from Prague. I think Uber ran me about $110 from Prague.
From the train/bus station, go toward town by going under the pedestrian underpass. That will put you out right by the Becherovka factory. (The English tours book out quick but would very much recommend their tour and tasting if you can score tickets on their website.) This is the beginning of a busy shopping street with knickknacks and inexpensive hotels but keep on walking, this is not the area you came to see. It will come to the Tepla river we’re you’ll cross and follow it to the heart of Karlovy Vary. Stroll through the colonnades, explore the iconic hotel lobbies, have a piece of famous Pupp cake at the GrandHotel Pupp, indulge in a spa treatment there. The only fault I see in your plan is not spending the night in this magical little time capsule.
