Can you tell us a little more about your interests? And maybe also how much of your time will be free (in other words do you have days before or after the conference or are you at the conference all morning or all day and want to explore each day after your conference sessions)?

I guess what I'm not understanding is why you're planning to make day trips to other countries instead of staying around to explore Prague which has so much to see!

Take a day trip by train to Dresden. It was probably the most beautiful city in Europe until the end of World War II when Allied bombing occurred. It's just being restored, and museums are incredible.
You could also continue by train into Berlin from Dresden.
Another good day trip would be to Cesky Krumlov--about 2 hours south of Prague by bus.
Vienna is about 3 hours by train--a great large city with great history.

Add one hour to David's times. Prague - Cesky Krumlov 3 hours. Prague - Vienna 4 hours. The only foreign city reasonable for a day trip from Prague is Dresden (about 2 hours). But I agree with others - there is a lot of to see in Prague and its surroundings. You don't have to be spending most of your day on train or bus. Instead of trying to suggest what you could see I strongly recommend Rick Steves' book Prague & the Czech Republic. All major sights of Prague and C.R. are described there.

For a day trip only Germany is close enough to Prague. It takes 2 hours (by direct train) to get to Dresden. The old town was completely leveled in WW2, but the most important buildings were reconstructed. And the city has world-class museums, among them a armory, the Old Masters, and the Green Vault(s). BTW: the Green Vault has a exhibit called "The Royal Household at Delhi on the Occasion of the Birthday of the Grand Mogul Aureng-Zeb".
Also on the route to Dresden is Saxon Switzerland, which are Central Europes most picturesque mountains.
You could also see a smaller town like Pirna (on the direct train route, but with a change), Meissen, or Freiberg (train change in Dresden).

At https://www.ricksteves.com/europe/czech-republic, the At a Glance list gives you an idea of the towns covered in Rick Steves' Prague and Czech Republic guidebook (available through Amazon.com for their Kindle app), as well a map of the country. The thin black lines on that map are the train lines, so you see a number of towns and castles highlighted that are served by train. Castles visits are usually with a guided tour that you book upon arrival. The Terezin Concentration Camp memorial is more conveniently visited by regular bus or with a tour departing from Prague.

Looking Up Train Schedules and Routes Online gives you the Deutsche Bahn train schedule link and tips for using it. You can also see train lines and stations on Google Maps, but you have to zoom in a lot.

Other worthwhile destinations within easy reach of Prague are Tabor, Hradec Kralove and Plzen.
Terezin is a pleasant 30 mins walk along the river from the railway station in Jeromerice.

Even closer to Prague - Melnik - historical wine town on the confluence of rivers Vltava and Labe (Elbe).

From the castle in Melnik (1 hr from Prague) you have a wonderful view. Enjoy it and continue your journey to Litomerice, 30 mins away on the same railway line.
