We are staying 5 nights in Dubrovnik in September and would like to then travel (by bus?) to Peljesac and spend a couple of nights there. We will want to hike/walk, visit wineries, see the local people and culture etc.

You could catch a bus to Ston or Orebic, but I think you will struggle to get the most out of the area without a car.
