This will be our 13th RS Tour and my spectacular sister-in-law is on the waitlist. Wondering if any single females would be open to sharing a room? My SIL is 60, single, an architect, and a wonderful travel buddy. This will be her first RS tour but we promise to train her LOL

Did you check to see if there are even any spots left? If the tour already has the max number of participants (28), they can’t just add more people.
For what it is worth, I have gotten on a tour from the waitlist. In my case, it happened just before the 60-day final payment deadline. I hope a single spot opens up for your SIL.
Many people who take RS tours don't know about this forum, but I hope you are successful getting a response. And just to add to Laura's response, I'd suggest calling the tour office to ask if there are any singles signed up for that specific tour. Even if there are cancelations that open up the wait list, if there are no other solos there is no opportunity for room sharing. As the single supplement is mandatory I don't know how the RS folks would handle a room share situation after one person originally signed up as a solo and paid the supplement (it is due with the final payment 60 days prior to the tour). Not to rain on your parade, just noting there may be some logistical challenges, but it's kind of you to look out for your sister in law. I hope she gets to join you!
