We will be leaving from the Port of Miami in April, 2027.
Flight schedules for us, to arrive the morning of departure, would require 12 hour red-eye flights, which we do not want.
So, we will fly in the day before and stay one night in a hotel - hopefully near the port.
Do any of the hotels near the port offer shuttles to the port? If not, how expensive are taxis in Miami?

I take it you are referring to a cruise? I would contact the cruise company for their advice. I am sure you aren't the first cruiser to want this information.

We paid around $30 for UBER from airport to the port of Miami. So depending on how near your hotel is to the port, it should be less than that.

You can check on Booking.com and then book directly with the hotel. Use Port of Miami as your location, then filter for price, amenities, etc as well as distance. There are hundreds within 5 km. Many have shuttles to the piers for a fee,but taxis aren't that expensive if you are staying close by. We stayed at the Courtyard by Marriott in the Brickell area.

South Beach has a lot of hotel choices and a great place to recover from jet leg. We just use Uber. to get to the port. I don't remember the cost but the fact that I don't remembers implies that it was reasonable probably less than $30.

I would stay in Brickell, it's a couple of miles from the port, the Uber would be about $20 to $30, it's a nice area to walk with lots of restaurants.
