We will be in Innsbruck for three days in late June 2018. Is it possible to get a day-long, round trip bus tour from there to Neuschwanstein castle area? The train routes look very difficult.

I did just want to mention that of the various places I have looked forward to seeing throughout the years, this one disappointed the most. Your time actually in the castle is quite short and the time and effort it took to get there for such a short time didn't seem like a good use of time to me. (I went on a day trip).
Everyone has their own tastes, but it may be that going to view this fake castle isn't worth the effort from Innsbruck.

I used Innsbruck Top Travel in 2014 for two private tours: (1) a nature day around Innsbruck and (2) a trip to Oberammergau and Neuschwanstein followed by a transfer to MUC airport. The owner -- Nacho Molina -- is one of the best guides I have encountered. He's passionate about showing Austria to visitors to Innsbruck. The group tours his company offers historically have been done in vans, not coaches, so it's a smaller experience.
As an aside, I loved Neuschwanstein, but my time there (including the standard tour inside the palace) was part of a day with an awesome guide that included much more than just Neuschwanstein.
